Synopsis for the March 20 episode of Supergirl, which is a prelude to the musical episode crossover with The Flash:
A new villain (guest star Teri Hatcher) comes to National City, putting Supergirl on high alert. Meanwhile, Winn’s alien girlfriend Lyra gets him in trouble with the law. Maggie attempts to help Winn but old loyalties get in the way. The Music Meister (Darren Criss) attacks Supergirl.

Odette Annable has been cast as Supergirl's season 3 main villain, Reign. She has previously appeared on various TV series, including House MD, Banshee, and The Astronauts Wives Club, as well as one of the lead characters in the film Cloverfield.

In the comics, Reign was one of 5 WorldKillers genetically engineered by a secret research initiative of the Kryptonians; Doomsday is allegedly one of them.
